Effect of quercetin on nonshivering thermogenesis of brown adipose tissue in high-fat diet-induced obese mice

چکیده

Activating nonshivering thermogenesis in brown adipose tissue (BAT) is a promising strategy to prevent obesity. This study investigated whether quercetin supplementation improves obesity mice by increasing BAT and white (WAT) browning. Compared high-fat diet (HFD)-fed mice, fed HFD supplemented with 1% (HFDQ) had reduced body weight total plasma cholesterol. In HFDQ-fed retroperitoneal WAT (RWAT) was decreased, browning effect lipolysis were increased. increased expression of genes BAT, including uncoupling protein 1 (UCP1), peroxisome proliferator-activated receptor-gamma coactivator 1? (PGC1?), cell death-inducing DFFA-like effector A (CIDEA), mitochondrial transcriptional factor (mtTFA). Quercetin proteins ?3-adrenergic receptor (ADRB3), p38 mitogen-activated kinase (MAPK), AMP-activated (AMPK) pathways HFD-fed which suppressed an AMPK inhibitor or ADRB3 antagonist. Energy expenditure core temperature not changed quercetin, but physical activity HFDQ during dark periods at room cold temperatures. also decreased the Firmicutes Bacteroidetes ratio short-chain fatty acid production feces mice. summary, may attenuate Although did show consistency data molecular pathophysiological levels between function obesity, it shows health effects accompanied improved gut microbiota dysbiosis.

NT-PGC-1α Activation Attenuates High-Fat Diet–Induced Obesity by Enhancing Brown Fat Thermogenesis and Adipose Tissue Oxidative Metabolism

The transcriptional coactivator peroxisome proliferator-activated receptor γ coactivator (PGC)-1α and its splice variant N terminal (NT)-PGC-1α regulate adaptive thermogenesis by transcriptional induction of thermogenic and mitochondrial genes involved in energy metabolism. We previously reported that full-length PGC-1α (FL-PGC-1α) is dispensable for cold-induced nonshivering thermogenesis in F...

Reduced brown adipose tissue thermogenesis of obese rats after ovariectomy.

Brown adipose tissue (BAT) thermogenesis was assessed by measuring mitochondrial guanosine diphosphate (GDP) binding, cytochrome oxidase activity and oxygen consumption in ovariectomized (OVX) and sham-operated rats. The food intake and body weight of OVX rats increased more than those of controls and OVX rats became obese. Mitochondrial GDP binding, as an indicator of thermogenic activity, cyt...

Effects of epigallocatechin-3-gallate on thermogenesis and mitochondrial biogenesis in brown adipose tissues of diet-induced obese mice

Background: Epigallocatechin-3-gallate (EGCG) is the major polyphenol in green tea and has been considered a natural agent that can help to reduce the risk of obesity. Objective: The aim of this study was to investigate the effects of EGCG on thermogenesis and mitochondrial biogenesis in brown adipose tissue (BAT) of diet-induced obese mice. Methods: Male C57BL/6J mice were provided a high-fat ...
